چکیده

In previous studies in this laboratory, highly purified liver microsomal cytochrome P-450 was shown to catalyze the hydroperoxide-dependent hydroxylation of a variety of substrates in the absence of NADPH, NADPH-cytochrome P-450 reductase, and molecular oxygen, and evidence was obtained that the oxygen atom in the product was derived from the peroxide. To determine whether the cytochrome functions in such reactions by a peroxidase-type mechanism, the kinetics of its interactions with a variety of substituted hydroperoxides and peroxy acids have been determined by stopped flow spectrophotometry. The reaction of P-450m, with various peroxy compounds yields an intermediate with an absorption maximum at about 436 nm in the difference spectrum, with pseudo-first order or biphasic kinetics depending upon the individual rate constants and the concentration of the oxidant used. The results are in accord with a reversible two-step mechanism, as follows: P-450 + oxidant e C + D,

Cytochrome C and Caspase-3/7 are Involved in Mycophenolic Acid-induced Apoptosis in Genetically Engineered PC12 Neuronal Cells Expressing the p53 Gene

Mycophenolic acid (MPA) is the active metabolite of mycophenolate mofetil. This study designed to investigate the mechanism of cytotoxicity of MPA on the genetically engineered PC12 Tet Off (PTO) neuronal cells with p53 gene. Alamar Blue (AB) reduction showed concentration-dependent cytotoxicity of MPA on PTO cells with IC50 value of 32.32 ± 4.61 mM. The reactive oxygen species (ROS) generation...

Efficacy of different antiemetics with different mechanism of action on xylazine induced emesis in cats

Xylazine hydrochloride, α2 adrenoceptor agonist, is an analgesic, sedative, tranquilizer, and muscle relaxant agent in veterinary medicine which is mostly used as in pre-anesthesia in veterinary medicine. One of the main adverse effects of xylazine which limits application of this medication in small animal veterinary practice (mostly in cats) is nausea and vomiting which can end up with aspira...

The Effect of Eight Weeks of Resistance Training with Tribulus Terrestris Extract on the Apoptosis Mechanism in the Heart Tissue of Rats Exposed To Stanozolol

Background: Artificial intelligence analysis based on the gene list obtained from the DisGeNET database identified the important genes involved in the heart damage process. Data enrichment highlighted the apoptosis signaling pathway as a vital pathway in cardiovascular risk. Hence, we estimated the binding affinity of chemical and plant bioactive molecules for cytochrome-c protein. Here, we per...

Novel Colchicine Analogues Target Mitochondrial PT Pores Using Free Tubulins and Induce ROS-Mediated Apoptosis in Cancerous Lymphocytes

B-acute lymphoblastic leukemia (B-ALL) is the frequent pediatric malignity. Chemotherapy is the most practical approaches to deal with such malignancies. Microtubule-targeted agents are one of the most strategic drugs which formerly use in chemotherapy.Although,colchicine-binding anti-tubulin agents exhibited promising effects in clinical trials, their exact mechanism of action is not fully und...
